Berberine is a naturally-occurring isoquinoline alkaloid isolated from Berberis species of plants, among others. Like many ingredients used in Traditional Chinese Medicine (TCM) for millenia, berberine is currently being studied for a variety of modern medicinal purposes.
Berberine's varied positive effects on many metabolic outcomes make it particularly of interest in treating the modern obesity-related condition known as "metabolic disorder". Unfortunately, bioavailability for this compound is low - estimated at less than 5% of the orally-administered dose - the compound itself being poorly soluble, and the gram-plus doses necessary for a physiologically-relevant effect cause major gastrointestinal side-effects. This is what many companies are NOT telling you. Super-Berberine™ solves these problems in two ways.
Firstly, in Super-Berberine™ the berberine is complexed with cyclodextrins. Cyclodextrins are sugar molecules bound into a ring- or doughnut-shape, which can be used to increase the solubility of other compounds by the formation of inclusion complexes. The insoluble compound is held in the hydrophobic cavity, and the cyclodextrin acts as a "carrier" molecule.
Berberine-cyclodextrin complexes are not only more soluble, and therefore more bioavailable, than plain berberine, but also sweeter and less bitter due to the natural sugary taste of cyclodextrins and because complexed molecules cannot fully interact with taste buds. This helps with glucose uptake into skeletal muscle likely via AMPK activation. The concern about AMPK causing muscle atrophy or inhibiting MPS is not playing out in human clinical settings, despite some theoretical concern based on in vitro studies. Ultimately, if someone is engaged in regular resistance training and consuming a protein adequate (> or = to 1.3g per lb bodyweight), with sufficient leucine "triggers" spread and distributed multiple times throughout the day, then there will be plenty of mTORC1 stimulation, along with PGC-1a. In this regard, AMPK is not the muscle protein "shredding" agent that many people have feared. Why? Because, as we explain above it does not occur in a vacuum, or in isolation. Ultimately, AMPK activation in the presence of intense exercise, anabolic nutrition (protein/leucine & optimal EFAs) provides a permissive opportunity to enhance body composition via both mitochondrial biogenesis, improved insulin sensitivity, and a "primed" fat burning environment. All, while not interfering with muscle protein accretion/ maintenance and muscle hypertrophy! The main limitation of absorption from the gut is ameliorated by capric acid (MCTs/ Coconut Oil, or Sodium Caprate). It helps to inhibit a p-glycoprotein that would otherwise reduce its systemic absorption.

Structure of sodium caprate
Sodium caprate is an absorption promoter, increasing the oral bioavailability of poorly permeable compounds across the gut wall. Many bioavailability enhancers work by increasing cellular permeability, allowing greater transcellular movement (through the epithelial cells) by diffusion or active transport. Sodium caprate dilates tight junctions (TJs) in the intestinal epithelium, increasing their permeability, facilitating paracellular transport (between the cells). Sodium caprate also inhibits the excretion pump function of P-glycoprotein, of which berberine is a substrate. As a sodium salt of a medium chain fatty acid, sodium caprate is Generally Recognized As Safe (GRAS) as a food additive. When sodium caprate is co-administered with berberine, intestinal absorption of berberine increases significantly, leading to bioavailability improvements of 1.5 to 2.3-fold.

B) Aframomum melegueta (75 mg) stnd to 12.5% 6-paradol) - [or High Quality Ginger Extract standardized for bioactives at 30%, and 6-,8-, and 10-gingerols/ shogaols at 20%]
Known more commonly as “Grains of Paradise” (GOP) is related to family of ginger spices (Zingiberaceae). GOP extracts possesses some intriguing properties that adds another rational pathway by which PARTITION-MD may effectively ‘coax’ human physiology into shuttling energy preferentially toward lean tissues (muscle), and away from adipose (fat). The bioactive pungent compounds 6-paradol, 6-gingerol and 6-shogaol appear to activate brown adipose tissue (rich in mitochondria), which is like the ‘ying’ to the ‘yang’ of the typical white adipose tissue that we all know and hate. While white adipose tissue stores excess calories like a storage closet, brown fat oxidizes excess calories to generate heat and burn fat rather than storing it. A single-blind, placebo-controlled 2013 study in humans demonstrated GOP extract supplementation increased energy expenditure and brown fat activation after a 4-week period of supplementation. In theory, the longer one supplements with GOP extract, the more responsive brown adipose tissue becomes. Hence, think about GOP as another tool in the PARTITION-MD armamentarium to help increase the size of the muscular compar™ent “storage tank,” while keeping a lid on fat cells, and limiting “spillover” into the adipose tissue compartment.
2. TO PROTECT KEY MOLECULES SUCH AS INSULIN RECEPTORS AND GLUT2 AND GLUT4.
GLUT2 transports glucose from the liver to the blood. GLUT4 helps to transport glucose to adipose and muscle tissues. Having these molecules in perfect working condition is key in nutrient partitioning. Think efficient clearing of glucose and you are on track with this.
CinnulinPF® (250 mg) is a patented water-soluble cinnamon extract, standardized to contain specific bioactive compounds called doubly linked procyanidin type-A polymers. This is important, because normal “spice rack” cinnamon varieties present in most dietary supplements can contain volatile oils that can be toxic. 4. IMPROVE GLUCOSE CONTROL AND INSULIN SENSITIVITY.
R-Alpha Lipoic Acid (100 mg) is a vitamin-like fatty acid found naturally in red meat, spinach, broccoli, potatoes, yams, carrots, beets, and yeast that is known as the “universal antioxidant”. This nickname stems from the fact that alpha lipoic acid is both fat and water soluble, and can therefore counteract oxidative stressors found inside the cell (i.e. as an intracellular antioxidant) as well as outside the cell membrane (i.e. as an extracellular antioxidant). Alpha lipoic acid is found in every cell in the body, serving as a coenzyme that controls carbohydrate metabolism as well as the production of adenosine triphosphate (ATP) from the citric acid cycle. Another highly unique quality of alpha lipoic acid is that it can regenerate endogenous antioxidants, such as vitamin E, vitamin C, and glutathione. Other important roles are metal chelation (i.e. binds to and helps eliminate copper, lead, zinc, mercury, iron), brain health, blood vessel function, neuronal conduction velocity, and (of course) its well-known effects on promoting insulin sensitivity and glucose disposal.
Momordica charantia/Bitter Melon (50 mg) is a vegetable grown in parts of Asia, Latin and South America that is affectionately known as “vegetable insulin” for its beneficial effects on glucose control and insulin signaling. Mormordica charantia contains over 20 active constituents including an insulin-like polypeptide called “polypeptide P” and as been shown to increase pancreatic insulin secretion, hemoglobin levels, red blood cell concentrations, and hepatic (liver) glycogen synthesis. It also decreases hepatic gluconeogenesis and may increase AMPK signaling for enhancing metabolic function--an effect that is similar to Metformin and may be useful for fat loss.
Chromium Picolinate (200 mcg) is a combination of chromium (an essential trace mineral) and picolinic acid (a naturally occurring chelating agent the body uses to absorb and transport certain minerals). Chromium helps regulate blood sugar and insulin sensitivity, improve mood, and reduce appetite (especially binge eating of carbohydrates). Like most bioactive ingredients, these effects are all dose related (i.e. are more consistently observed when higher doses are used) and tend to vary among individuals. Biotin (2.5mg) is a vitamin that is found in small amounts in numerous foods. Biotin has been shown to play a key biochemical and physiological role in metabolic function, nutrient and glycemic uptake into skeletal muscle. Moreover, the co-administration of chromium with biotin appear to interact to positively influence insulin sensitivity in various preclinical and clinical models. Put simply, human studies show that biotin helps chromium work better.
Bioperine (5 mg) is a patented form of black pepper extract containing the active ingredient piperine.
